The affinities for asymmetric conformations of calmodulin were estimated based on assumptions and actual affinity measurements from reference [7]. The affinity of WFF for the RR state was only given as upper limit (<0.1nM), and was assumed to be 0.05nM. The affinity of both peptides for the TR state was assumed to be equal to the peptide affinity for the isolated C-lobe in the R state. In the case of WF10, which is supposed to only interact with the C-lobe, the peptide affinity for the RR and TR states were set to be equal. Each peptide’s affinity for the RT conformation (which is expected to be underpopulated in steady-state conditions and therefore play a marginal role in this analysis) was set equal to the affinity for the TT state.
