Activation of mast cells through FcεRI can result in allergic responses. The identity of mast cells progenitors is controversial, in particular whether these cells express FcεRI. Here, we performed single-cell RNA sequencing of human peripheral blood progenitors to study the appearance of FcεRI during hematopoietic progenitor differentiation. We performed single-cell RNA sequencing of FACS isolated human hematopoietic progenitor cells. The first dataset constitutes Lin− c-Kit+ cells sorted from a peripheral blood sample. The second dataset constitutes Lin− CD34+ c-Kit+ cells with spike-in Lin− CD34+ c-Kit+ FcεRI+ cells. Briefly, to generate the latter dataset peripheral blood cells from a donor were divided into two fractions. Cells from one fraction was labeled with phycoerythrin (PE)-hashtag-oligonucleotides. Lin− CD34+ c-Kit+ cells were sorted from the unlabeled fraction and Lin− CD34+ c-Kit+ FcεRI+ cells were sorted from the PE-hashtag-oligonucleotide-labeled fraction. The two sorted cell fractions were pooled and single-cell transcriptomics was performed. **As Swedish legislation and GDPR prevent unrestricted distribution of raw sequencing data, raw sequence data cannot be provided, including through controlled access databases.
