To demonstrate the use of the synthetic DNA ladder spike-ins in genome sequencing, we added the ladder into well-characterised reference human DNA samples (NA12878, NA24385, NA24149 and NA24143) and performed whole genome sequencing. We show how the ladder can be used as a reference to estimate technical variation and measure quantitative genetic features within and between samples.
