In this study we aimed to find the differences between several CAS systems: CAS9, CRISPRa, CasRx and hfCasRx. For that we created libraries of gRNA sequences targeting different genes and cloned them to be compatible with the 10x Genomics scRNA-seq pipeline. scCRISPR screens were performed in MDA-MB-231 cells with two replicates per system. CRISPR-Cas9 samples were sequenced at day 17 after induction; CRISPRa and hfCasRx samples were sequenced at day 12, while one of the CasRx samples was sequenced at day 4 and the second one at day 12.
