We use in vivo mouse models to characterize the outcome of deleting PI5P4Ka in prostate basal cells. Single cell RNA-Seq analysis reveals the molecular consequence of altering PI5P4Ka and links impaired lipid metabolism with delayed mPIN development. Overall design: We used a flow cytometry-based selection of eYFP+ cells from our luminal and basal in vivo systems. Two animals per group and 1500 individual cells were sequenced following FACs selection using the SORT-seq RNA-Seq platform, a partially robotized method based on the CEL-Seq2 protocol.
