An RNA binding protein RBPm1 regulates the axoneme assembly in male gametocytes of Plasmodium yoelii. To understand the molecular mechanisms underlying this process, we performed RNA-Seq to examine the male transcriptome changes due to the loss of RBPm1. We used a DFsc7 reporter strain, which specifically expressed GFP in the male gametocytes, to obtain high-purity male gametocytes. We deleted the Rbpm1 in the DFsc7 strain and generated a mutant strain. Male gametocytes were sorted from DFsc7 and mutant strains by fluorescence-activated cell sorting (FACS) according to GFP fluorescence intensity and subjected to RNA-seq.
