Investigation of the mechanism by which combined EZH2 and HDAC inhibition kills PC3 prostate cancer cells. Next-generation sequencing was used to identify changes in genome localization of HDAC targets (H3K27ac) and EZH2 targets (H3K27me3) upon treatment with each inhibitor alone or in combination. Overall design: Examination of genome-wide localization of 2 different histone modifications in PC3 prostate cancer cells following treatment with Vehicle (DMSO), HDAC inhibition (VOR, Vorinostat), EZH2 inhibition (GSK126), or combined HDAC and EZH2 inhibition (COMBO)
