Single cell RNA sequencing reveals the aging associated change of ovarian macrophage populations

Macrophages are important modulators of the ovarian tissue microenvironment, and are associated with the ovarian aging process. To dissect the changes of macrophages during ovarian aging, we used a combination of single cell RNA sequencing (scRNAseq) and flow cytometry to inspect the CD45+ CD11b+ myeloid cells in young (3 months old) and aged (15 months old) murine ovaries, which mainly consists of macrophages, monocytes and dendritic cells. CD45+Cd11b+ cells were isolated from 3 months old or 15 months old murine ovarian tissue using fluorescence assisted cell sorting (FACS). For the each age group, 3 biological replicates were included, each consists of cells harvested from two virgin C57BL/6J mice and was tagged with TotalSeq B series hashtag. The replicates from each age group were pooled before sorting and library preparation.
