¹H NMR spectra of raw cow milk before and after centrifugation-induced somatic cell count reduction

In this dataset, we provide ¹H NMR spectra of bovine raw milk samples with varying somatic cell count (SCC) levels before and after artificial centrifugation for SCC reduction. Milk samples were collected from four Czech dairy herds during 2023–2024 and represent Holstein and Czech Fleckvieh breeds. A total of 56 milk samples were collected, each measured twice: prior to centrifugation (BEFORE) and after centrifugation (AFTER), resulting in 112 ¹H NMR spectra. Samples cover a wide SCC range from below 200 × 10³ cells/mL to values exceeding 400 × 10³ cells/mL. High-SCC samples were prepared by mixing bulk tank milk with mastitis milk from untreated cows. Milk fat was removed by centrifugation and milk plasma was extracted. Proteins were precipitated with methanol, evaporated, and reconstituted in water with phosphate buffer containing TSP as an internal chemical shift reference. Spectra were acquired on a Bruker Avance III HD 500 MHz spectrometer using a NOESY-presaturation pulse sequence at 298 K. The resulting spectra were automatically phase-corrected (zero- and first-order). In addition to spectral data, the dataset also provides supplementary sample information, including average SCC, pH value, milk freezing point, lactose content, and electrical conductivity of milk for each analyzed sample.
