### README – PBMC Transfection and Caspase Assay Data **Project Overview** This dataset presents raw and processed data obtained from peripheral blood mononuclear cells (PBMCs) transfected with miRNA-133b and subsequently infected with SARS-CoV-2. The data include results from cell viability assays, GFP-based transfection efficiency measurements, and caspase-1 luminescence assays. Each assay includes both raw experimental outputs and processed sheets with percentage calculations and statistical testing. **Contents** - Cell viability data (PBS and RPMI washed)- Transfection efficiency (% GFP-positive cells)- Caspase-1 activation (RLU values, raw and blank-subtracted)- MOI-dependent infection data- Descriptive statistics and Mann–Whitney test results **Excel Sheet Overview** - `Viability_raw`: Total and dead cell counts - `Viability_final`: Percent viability with statistics - `Transfect_raw`: Total vs. GFP+ cell counts - `Transfect_final`: Transfection efficiency (%) - `GloCaspase_infected_raw`: RLU values, infection model - `GloCaspase_infected_transformed`: Normalized luminescence - `GloCaspase_miR_raw`: miRNA-related luminescence, raw - `GloCaspase_miR_transformed`: Blank-corrected, processed **Measurement Notes** - RLU = Relative Luminescence Units - GFP = Green Fluorescent Protein (transfection indicator) - All luminescence data were recorded using a multimode plate reader. **Contact** For queries related to dataset use or interpretation, please contact Johany C.
