Non-alcoholic fatty liver disease (NAFLD) is a metabolic disease with complex mechanisms involving numerous inducing factors, changes in various metabolic functions, and has a variety of animal models. Most NAFLD drugs, however, have shown inconsistent efficacy in preclinical and clinical trials, mainly due to the fact that preclinical models predominantly have different limitations and lack parallel assessments of similarity with NAFLD progression. The similarity matrix analysis shows that the High-Fat/High-Cholesterol/Drinking-Fructose-induced model (HFHC_F) and CCl4-induced model have higher similarity with NASH and NASH patients with advanced fibrosis, respectively, but HFHC_F is inconsistent in cholesterol and bile acid metabolism. To take advantage of each, we create a mixed model called High-Fat/High-Cholesterol/Drinking-Fructose with subsequent CCl4-induced model (HFHC_F_C) and evaluate changes. Our study proves that the HFHC_F_C model has great similarity with NASH patients with advanced fibrosis, especially in cholesterol and bile acid metabolism.
