This work describes the function and targets of a maternal KRAB-Zinc finger protein, ZFP708, which specifically mediates maintenance of DNA-methylation at a subset of LTR retrotransposons during embryonic epigenetic reprogramming. HA-/Flag-tagged ZFP708 or Flag-tagged ZFP708 lacking KRAB domain were overexpressed in mouse embryonic stem cells to identify genome-wide binding sites by ChIP-sequencing. In ZFP708 overexpressing cells TRIM28/Kap1 recruitment and H3K9me3 dynamics were also assed by ChIP-sequencing.
