In this project, data were collected with multiple microscopy techniques to longitudinally monitor the development of osteogenic microtissues on chip and perform correlative imaging. On-chip cultures were live stained and imaged with fluorescence microscopy (incl Airyscan module). After fluorescence imaging they were kept in culture and monitored at the next timepoint, until the timepoint of interest was reached. The samples were then extracted with an biopsy punch and cryopreserved by high-pressure freezing for sequential cryo-fluorescence microscopy and volume electron microscopy. Additionally, chemical characterizations were performed with Raman spectroscopy. Each chip contained maximally 5 microtissues in separate chambers (Cav1-5). The aim of this study was to develop a device that is suitable for whole-tissue imaging and subsequent correlative imaging. This repository contains the data that were used for the manuscript "A workflow integrating organ-on-chip culture and correlative 3D light and electron microscopy for microtissue analysis".
