We mapped the genome-wide binding profiles of BMAL1 and REV-ERB⍺ during peak protein expression of each factor (ZT4, and ZT8, respectively) by ChIP-Seq in gastrocnemius muscles from control C57BL/6J mice. Wildtype 5 month old male C57BL/6J mice maintained on 12:12 light:dark and standard chow diet were sacrificed by cervical dislocation and skeletal muscle was immediately collected and snap frozen in liquid nitrogen. ChIP was performed as previously described (Uhlenhaut et al. 2012). Sheared chromatin was immunoprecipitated either with anti-BMAL1 antibody (ab93806; LOT: GR279846-2), a mixture of anti-REV-ERB⍺ (#23678 and #27401 produced by Ron Evans), or anti-RNA Polymerase II (Biolegends #MMS-126R; clone 8WG16). Two replicates of BMALl1, RNA polymerase 2, and Rev-erb⍺ were made, for a total of 6 ChIP-seq samples + 1 input.
