Identify VDR targets within the proximal and distal intestine using a genome-wide method.

The object of this aim is to understand the mechanism by which vitamin D mediates calcium absorption. Our working hypothesis is that VDR has distinct sets of binding sites: proximal versus distal intestine. Thus, to accomplish the goals, we will 1) determine whether 1,25(OH)2D3/VDR regulates specific target genes in proximal and distal intestine, 2) identify the locations of genomic binding sites for VDR in proximal and distal intestine. Overall design: Mouse data comprising RNA-Seq from 24 samples was obtained: 8 samples from the colon (4 vitamin D treated replicates, 4 vitamin D-deficient replicates), 8 samples from the duodenum (4 vitamin D treated replicates, 4 vitamin D-deficient replicates), and 8 samples from the ileum (4 vitamin D treated replicates , 4 vitamin D-deficient replicates).
