To test if CWC15 is required for miRNA accumulation, an artificial miRNA (amiRCWC15) targeting 5’ UTR (Untranslated region) was used to knockdown the transcript levels of CWC15. Next, to test the effect of CWC15 on miRNA accumulation at global levels, flower inflorescences of four-week old col-0 and amiRCWC15 were collected, and total RNA was extracted by Trizol for Illumina deep sequencing analyses of miRNA profile.
