16s sequence in DSS-induced colitis

This project focuses on 16S rRNA gene sequencing of colon tissue samples derived from C57BL/6 mice (6-8 weeks old) before and after dextran sulfate sodium (DSS)-induced colitis modeling. A total of 16 mice were divided into 4 experimental groups with 4 mice per group, following a randomized design:Wild-type (WT) group (without DSS treatment);WT + DSS group (WT mice subjected to DSS-induced colitis);MERTK knockout (MERTK-/-) group (MERTK gene-deficient mice without DSS treatment);MERTK-/- + DSS group (MERTK-/- mice subjected to DSS-induced colitis).The primary objective of this project is to analyze the dynamic changes in the composition, diversity, and relative abundance of the colonic microbiota across the four groups before and after DSS modeling. By comparing microbial community differences between WT and MERTK-/- mice, as well as between DSS-treated and non-treated subgroups, this study aims to explore the regulatory role of the MERTK gene in colonic microbiota homeostasis under inflammatory conditions (induced by DSS), and to provide foundational data for understanding the association between host genetic factors, intestinal microbiota, and DSS-induced colitis pathogenesis.
