C. elegans lacking AMPK do not exhibit reproductive defects during normal growth, but when passaged through dauer, exhibit germline hyperplasia and premature spermatogenesis, followed by complete sterility post-dauer (Narbonne and Roy, Development, 2006). Loss of a novel RNA helicase HZL-1 suppresses these phenotypes. We sought to carry out carry out a CLIP-Seq to determine what RNAs, if any, are bound by HZL-1 in the absence of AMPK in the dauer stage. CLIP-Seq of dauer (96 hours post-hatching at restricted temperature) in the daf-2 and daf-2; aak-1; aak-2 (aak(0)) and hzl-1 genetic background harboring a hzl-1::gfp transgene expressed under intestinal or endogenous promoter. After immunoprecipitation with anti-GFP, samples were treated with standard Trizol RNA extraction of collected samples.
