This project aims to identify conditionally essential genes in the epidemic Staphylococcus aureus USA300 JE2 strain using Transposon-Directed Insertion Site Sequencing (TraDIS) in murine infection models. The mutant library was screened in both a skin infection model, where abscesses were collected, and a systemic infection model, involving the kidney and spleen. For each organ, sequencing was performed on individual samples from two mice as well as pooled samples from eight mice, providing a comprehensive overview of tissue-specific gene fitness requirements.
