For information on the proper use of pre-publication data shared by the Wellcome Trust Sanger Institute please see http://www.sanger.ac.uk/datasharing/ Only a few genomes of phages infecting Campylobacter jejuni have been sequenced, due to two unusual features of the DNA. One obstacle is that the phage DNA contains novel unidentified modified CG bases that changes the biochemical nature of the DNA, making isolation and purification very difficult. Secondly, the phage genome contains up to five regions of each 0.5-3 kb of direct large repeats, making assembly of Illumina derived sequences impossible. We have developed methods for more efficient isolation and purification that will allow PacBio sequencing.
