Characterization of EVI1+ murine AML leukemia

KMT2A-MLLT1 AML mice model were generated by using EVI1-GFP-knockin mice, where EVI1-IRES-GFP allele was inserted into the MECOM locus to monitor Evi1 expression by GFP. GFPhigh and GFPlow L-GMPs were compared by RNA-seq. The effect of ERG and Cyclin D1 silencing was assessed by shRNA against ERG (shERG) and CCND1(shCCND1) using retrovirally EVI1-transformed murine AML bone marrow cells. To determine transcriptional target genes of EVI1, H3K4me3 ChIP-seq was performed using leukemia cells collected from EVI1-transformed murine AML bone marrow cells.
