NEK8 blockade potentiates radiation-induced antitumor immunity and abscopal effect via lactate metabolism reprogramming in colorectal cancer (PRJCA036988)

In this project, we established an intratumoral iteration model of colorectal cancer radiotherapy resistance, designated as IR-MC38, and compared its tumor microenvironment to that of the parental MC38 tumor using single-cell sequencing. Through RNA sequencing, we identified NEK8 as a key gene associated with the anti-tumor immunity of CD8+T cells. Subsequently, we performed comprehensive multi-omics analyses on NEK8, including proteomics, CUTTag sequencing, and bulk RNA sequencing, to elucidate its role and underlying mechanisms.
