Langerhans cells were prepared from skin or derived from cord blood CD34+ cells Epidermal sheets were digested with dispase. CD1a+ cells were isolated using Myltenyi beads. CD1a-depleted cells were used to control keratinocyte contamination. CD1a+ Langerhans cells were isolated using Myltenyi beads from human cord blood CD34+ cells purified using Myltenyi beads in the presence of recombinant TGFb, SCF, GM-CSF, FLT3.
