To identify genes regulated by FOXO1 in HDLEC, we have empolyed whole genome microarray expression profiling. We identified CXCR4 as the most reduced gene after 72h of siRNA treatment for Foxo1 and the expression change was confirmed by RT-PCR and western blot. Using total RNA extracted from cultured HDLEC at 72h after transfection with siRNA mix targetting for Foxo1 and control mix , we measured gene expression changes. A mixed HDLEC derived from 3 male foreskins was used. Expression changes were confirmed by RT-PCR or WB.
