This study investigates the de novo generation and accumulation of copy-back viral genomes (cbVGs) during Sendai virus infection. Using Sendai virus strain Cantell as the parental strain, six independent recombinant viruses (rSeV C-eGFP) were rescued to generate Passage 1 stocks. Four of these P1 stocks were serially passaged at high multiplicity of infection, and viral RNA from the Passage 3 and 8 virus stocks was sequenced. The resulting data provide insights into the diversity, reproducibility, and evolutionary dynamics of cbVG formation across biological replicates and passages.
