TCR sequencing of alpha-beta and gamma-delta tumor infiltrating lymphocytes

Bulk and gamma-delta enriched tumor infiltrating lymphocytes (TIL) were expanded from surgically resected pancreatic ductal adenocarcinoma (PDAC) tissue according to the rapid expansion protocol. High-throughput sequencing of all TCR chains (TRA, TRB, TRD, and TRG) was conducted using the iR-RepSeq+ service provided by iRepertoire, Inc. (Huntsville, AL, US). The goal was to characterize the diversity of the expanded TIL in relation to their functional activity.
