Aging is one of the most important risk factor for cancer. However, next to nothing is known about how the age of the cell of origin impacts the evolution of tumor cells. Here we interrogated the cell-intrinsic forces that drive the progression of aged vs young cell of origin for lung cancer by using ex vivo transformation approaches Normal type 2 alveolar epithelial cells from aged vs young Kras-LSL-G12D/+; Trp53 fl/fi; Rosa26-LSL-EGFP-Cas9 (KPCas9) mice were isolated by FACS and transformed ex vivo. These tumor cells were maintained in 3D culture (tumor organoid) for 8 passages and samples from each passage, including untransformed alveolar organoids.
