Fecal and serum samples were subjected to untargeted metabolomics analysis at Majorbio Bio-Pharma Technology Co., Ltd. (Shanghai, China) using the Thermo UHPLC-Q Exactive HF-X system equipped with an ACQUITY HSS T3 column (Waters, USA). Raw LC/MS data were initially processed using Progenesis QI software (Waters Corporation, Milford, USA). Metabolite identification was performed by searching the Human Metabolome Database (HMDB, http://www.hmdb.ca/), Metlin (https://metlin.scripps.edu/), and the Majorbio database. The resulting data matrix was uploaded to the Majorbio Cloud platform (https://cloud.majorbio.com) for further analysis. The Kruskal-Wallis test was used for statistical analysis, while partial least squares discriminant analysis (PLS-DA) was performed to assess metabolic differences. Pathway enrichment analysis was conducted using the Kyoto Encyclopedia of Genes and Genomes (KEGG) database (http://www.genome.jp/kegg/), mapping the identified differential metabolites to their respective biochemical pathways. Metabolic set enrichment analysis (MSEA) was conducted using the MetaboAnalystR package. The concentration change patterns of all identified metabolites were analyzed using the Small Molecule Pathway Database (SMPDB) through the Over-Representation Analysis (ORA) mode of MSEA.
