This dataset contains replication data for the manuscript: “Development of a three-dimensional vitrification protocol for domestic cat cumulus-oocyte complexes and comparison with standard vitrification”. Sodium alginate was employed as a matrix for the encapsulation of cat cumulus-oocyte complexes. Four experiments were performed to assess: 1) the optimal concentration of alginate; 2) the permeability of alginate to cryoprotectants used for vitrification; 3) the effect of vitrification in the alginate matrix on oocyte morpho-functionality and 4) the effect of vitrification in the alginate matrix on oocyte development into embryos in vitro. Data of each experiment consists of: 1) viability, nuclear status and cumulus expansion of fresh cumulus-oocyte complexes encapsulated in alginate at different concentrations; 2) concentration of the cryoprotectants dimethyl sulfoxide (DMSO) and ethylene glycol (EG) in fresh cumulus-oocyte complexes after exposure in standard conditions or after encapsulation in sodium alginate; bovine cumulus-oocyte complexes were used as a model considering the availability of biological material; 3) viability, cytoskeletal integrity and nuclear status of cumulus-oocyte complexes following standard vitrification or vitrification in the alginate matrix; 4) embryo development of cumulus-oocyte complexes following standard vitrification or vitrification in the alginate matrix.
