A M13 phage peptide display library containing 20 million independent phages was constructed using random oligonucleotides. Randomized peptide inserts were 9 amino acids in length and flanked by 4 glycines on each side. DR1-specific enrichment was performed by 4 rounds of selection (elution) and amplification (E.coli). Peptide M13P1-6 was selected at random from the DR-specific phage pool. Phage generated peptide M13P1-6 was able to out compete HA 307-319 for binding to DR1 molecules.
