Hippo Pathway Regulates Microarchitecture of Fibroblastic Reticular Cells in Lymph Node

To unveil the role of Hippo signaling in regulating the maintenance and integrity of FRCs in LNs. FRCs were sorted from models deleting Yap/Taz to inhibit hippo pathway, and deleting Lats1,2 to activate hippo pathway in FRC specific manner. We generated [1] FRC-driven Lats1/2 depleted mouse by crossing the PDGFRβ-Cre-ERT2 mouse with Lats1(flox/flox)/Lats2(flox/flox) mouse [2] FRC-driven Yap/Taz depleted mouse by crossing the FRC-specific CCL19-Cre mouse and the double Yap(flox/flox)/Taz (flox/flox) mouse. Lgr5+ stem cells in dermis were FACS-purified and used for ATAC-seq to explore Yap/Taz driven changes in chromatin accessibility
