In order to study the role of the transcription factor YY1 in oral cancer progression, transcriptome wide gene expression analysis was performed with inducible silencing of YY1 in a grade III, tongue derived oral cancer cell line, AW8507 (Established and characterized by Tatake RJ et al. 1990) stable for tet-ON-shYY1. AW8507 cells were transfected with TRIPZ-tet-ON-shYY1 plasmid and tet-ON-shYY1 stable cells were obtained by selection with puromycin followed by FACS for RFP positivity. The AW8507 cells stable for tet-ON-shYY1 were treated with doxycycline to induce tet-ON system, therefore expression of shRNA against YY1, which resulted in silencing of YY1 mRNA (Confirmed by qRT-PCR). Total RNA was isolated from control and doxycycline group and transcriptome wide gene expression changes were analyzed via microarray based approach.
