A novel affinity-tuned dual-checkpoint bispecific antibody with potent PD-L1 and moderate CD47 affinity was designed to improve the selectivity to TME and thus enhance antitumor immunity and efficacy. The gene expression analysis using Nanostring platform was performed to examine the immune cell modulation in TME following the CD47/PD-L1 bispecific treatment. MC38 tumor-bearing C57/BL6 mice were treated with vehicle control, aCD47, aPD-L1 monotherapy, their combination, or the bispecific antibody every 3 to 4 days starting at day 10 post-tumor implant. Tumors were harvested at day 20 or day 24 and preared for gene expression analysis by Nanostring platform (n = 7~9 mice/group).
