我们主要是通过荧光显微图片、电子显微镜图片和免疫共沉淀等实验数据,证实收缩丝体的存在以及收缩丝体与其他胞外囊泡的异同。首先,通过荧光显微镜和电子显微镜图片,我们观察到了收缩丝体会断裂成小珠子一样的囊泡。随后,我们提取了不同的胞外囊泡,并比较了各种囊泡所含蛋白的异同,发现收缩丝体与迁移体在蛋白组成上比较类似。该数据集的数据均为项目期间产生的数据,历时4年,该数据和论文于今年5月在杂志刊登,图片数据来源于我们的显微镜图片,所用的显微镜主要有尼康共聚焦显微镜、奥林巴斯共聚焦显微镜、扫描电镜等,所用的其他设备还包括电泳设备、转印设备和显影设备。
We primarily utilized experimental data including fluorescence microscopy images, electron microscopy images, and co-immunoprecipitation assays to validate the existence of contractosomes, as well as the similarities and distinctions between contractosomes and other extracellular vesicles. First, via fluorescence and electron microscopy imaging, we observed that contractosomes fragment into bead-like vesicles. Subsequently, we isolated distinct subtypes of extracellular vesicles, compared their respective protein compositions, and revealed that contractosomes exhibit highly similar protein profiles to migrasomes. All data within this dataset were generated over a 4-year project timeframe. This dataset and the accompanying manuscript were published in a peer-reviewed journal in May of this year. The microscopy image data were acquired using our laboratory's imaging platforms, primarily including Nikon confocal microscopes, Olympus confocal microscopes, and scanning electron microscopes (SEM). Additional experimental equipment utilized included electrophoresis apparatuses, western blot transfer systems, and chemiluminescence imaging development equipment.
