ATAC-seq of tobacco callus cells cultured by different NAA concentrations during in vitro tissue culture illustrates the landscape of chromatin openness across the entire genome,which provides a novel direction and theoretical basis for improving gene editing efficiency in plants by modulating chromatin accessibility through auxin signaling and tissue culture conditions. Approximately 80 tobacco explants were harvested from the media containing 0.1 mg/L or 0.5 mg/L NAA after 2 weeks culture for nuclei isolation. The ATAC-seq was performed according to the guidelines of the ATAC-Seq Library Prep Kit for Illumina®
