A number of mock zooplankton communities were constructed in order to validate species richness estimates generated by metabarcoding studies. These communities contained either a single individual or 'populations' of a number of different species, some of which were amplified with tagged primers. The communities were amplified at V4 region of the 18S gene and 454 pyrosequenced.
