After the glioma cells were digested with trypsin, 1×10⁶cells were taken in a 15mL centrifuge tube with a sharp bottom, washed twice with precooled PBS, and then cooled overnight in 70% ethanol. According to the instructions of cell cycle detection kit (Beyotime, Shanghai, China), propidium iodide staining was carried out, and flow cytometry (BD Accuri C6 Plus) was used for analysis. Results from cell cycle analysis demonstrate that either knockdown or overexpression of ASIC2 significantly alters the cell cycle progression and proliferation rate of glioma cells. Specifically, distinct changes were observed in the distribution of cells across different cell cycle phases, accompanied by corresponding increases or decreases in the rate of cell proliferation. These findings suggest that ASIC2 likely plays an important role in regulating the cell cycle in glioma cells.
