Protein Tyrosine Phosphatase Non-receptor type 22 (PTPN22) is encoded by a major autoimmunity gene and is a known inhibitor of T cell receptor (TCR) signaling. PTPN22 Ser449 is a novel phosphorylation site. Here we used optimized coding sequences (CDS) of PTPN22 WT or S449A eukaryotic expression vectors with N-terminal mEGFP and electroporated into either J.Lck WT or J.Lck Y192F cells to analyze the relationship between PTPN22 Ser449 phosphorylaion and Lck Tyr192 phosphorylaion by CITE seq. Electroporated cells were stimulated by 0.5ug/ml antibodies aginst human CD3/CD28 for 12 h and stained by Totalseq anti-human CD69 antibody. mEGFP+ T cells were sorted out and subjected to CITE- seq.
