This study aims to investigate the relationship between the hypothalamus and age-related DNA methylation in mouse model. The objective of this sequencing work is to compare the hypothalamus (HT) with two other components of the limbic system in mice, the hippocampus (HC) and the olfactory bulb (OB), to understand how DNA methylation levels in these regions are similarly versus differently altered as animals evolve from young age to middle age. Overall design: DNA was isolated from the hypothalamus (HT), the hippocampus (HC), and the olfactory bulb (OB) of standard male C57BL/6 mice at a young age (2 months, 2M) vs. an old age (12 months, 12M). This experiment contained 3 mice per age group, which are named Young1-3 and Old1-3. DNA libraries were prepared and then processed through bisulfite oligonucleotide-captured sequencing (BOCS) procedure. Sequencing was done with an Illumina MiSeq PE75 for quality control and with NextSeq500 High PE75 for obtaining DNA methylation profiles.
