CytoCV v2.0.0 Stu2 nuclear-to-cytoplasmic intensity dataset

This dataset contains the processed per-cell measurements and representative microscopy image data underlying the Stu2 nuclear-to-cytoplasmic intensity illustrative analysis presented in the SoftwareX manuscript “CytoCV: A web-based computer vision platform for reproducible quantitative analysis of yeast microscopy images.”The dataset compares STU2 WT and stu2 KR/AA cell pairs analyzed using CytoCV v2.0.0. The supplied CSV contains the combined per-cell measurements for both experimental groups, with WT measurements followed by mutant measurements. The accompanying Excel workbook contains the same measurements organized into separate WT and mutant worksheets.The representative wild-type image used in the manuscript and supplementary video is source file 210615_M2208_008. Cell 3 is the representative cell pair shown in the manuscript and has a nuclear-to-cytoplasmic intensity ratio of 0.883; the corresponding WT group average reported during figure preparation was 0.849.The representative mutant image is source file 210615_M2210_008. Cell 5 is the representative cell pair shown in the manuscript and has a nuclear-to-cytoplasmic intensity ratio of 0.468; the corresponding mutant group average reported during figure preparation was 0.513.For the current CytoCV analysis, the DIC-derived retained cell-pair mask defines the whole-cell-pair measurement region. Nup2-mKATE is mapped to the Red role and defines the nuclear contour, and Stu2-GFP is mapped to the Green role and provides the fluorescence measurement signal. Nuclear contours are clipped to the retained DIC-derived cell-pair mask. Cytoplasmic intensity is calculated as whole-cell-pair Stu2-GFP intensity minus nuclear Stu2-GFP intensity, and the nuclear-to-cytoplasmic ratio is reported when the cytoplasmic denominator is positive and finite.The images were selected from the broader experimental image collection generated for the previously published Stu2 localization study by Abouelghar et al. The present dataset documents the image subset and CytoCV analysis used for the SoftwareX illustrative example and is not intended to reproduce the exact image selection or analysis execution used for the original published comparison.
