Higher order mutants between PXY and ER families are characterised by profound secondary growth defects. It is likely that genes involved in regulating cambium homeostasis are differentially expressed in these backgrounds and consequently they could be used as a tool for gene discovery. The secondary growth region of 3-week-old hypocotyls and roots from wild type, pxy pxl1 pxl2 (pxf), er erl2, and pxf er erl2 lines were analysed for tissue composition and subjected to RNA-seq. Overall design: RNA-seq profiling of 3-week-old Arabidopsis wild type (Col-0), pxf (pxy-3 pxl1-1 pxl2-1 triple mutant, Etchells et al., 2013), er erl2 (er-105 erl2-1 double mutant), and pxf er erl2 (quintuple mutant, Wang et al., 2019) lines. Hypocotyls and the upper 5mm of root were excised prior to RNA extraction. Comparisons allow the identification of differentially expressed genes in radial growth. Each genotype has three biological replicates.
