This project aims to address the lack of tumor targeting of fluorouracil in the clinic as well as the low efficiency of conversion to the active form. We synthesize nucleic acid aptamer-fluorouracil couplers that can both target and deliver fluorouracil to the tumor site and increase the efficiency of fluorouracil conversion to FdUMP, thereby increasing its antitumor activity and reducing its toxic side effects. We sequenced the RNA transcriptome of drug-treated MIA PaCa-2 cells to analyze the changes in the transcriptome after drug treatment, providing a theoretical basis for 9Fu-AS1411-targeted pancreatic cancer therapy.
