We adapted the Aldehyde reactive probe (ARP; O-(biotinylcarbazoylmethyl) hydroxylamine) pulldown method to map ALKBH1 dependent targets in a transcriptome-wide manner. Overall design: Fragmented total RNA extracted from WT and ALKBH1 knock out cells was first incubated with ARP and then enriched by streptavidin pulldown. RNA were subject to Iclip library construction and high throughput sequencing.
