This study explores how treating peripheral blood mononuclear cells (PBMCs) with IronQ, a complex of iron and quercetin, affects their gene expression profiles. PBMCs are a promising source of stem cells for regenerative medicine due to their accessibility and storage convenience. Previous research suggested that IronQ-treated PBMCs exhibit potent angiogenesis and wound-healing properties. Using RNA sequencing analysis, the study identified significant gene expression changes in PBMCs from healthy volunteers and those with diabetes mellitus (DM) treated with IronQ. These changes included the upregulation of genes associated with inflammatory responses, cell migration, stress responses, and angiogenesis. Moreover, a network analysis revealed 20 hub genes crucial for various cellular processes, including immune responses and tissue repair pathways. Overall, this research suggests that IronQ treatment can significantly alter gene expression profiles in PBMCs, activating pathways essential for tissue repair and immune response, particularly beneficial for diabetic wound healing.
