Convergence of distinct RNA-silencing pathways on GW182/Tnrc6 [sgRNA screen]

sgRNA screens for factors influencing Trim71 silencing activity. Screen1: Genome-wide screen: Cells were transduced with two separate lentiviral sgRNA libraries (library 1: 52,000 unique sgRNAs (~5 per gene); library 2: 39,000 unique sgRNAs (~5 per gene); (Zeng et al., 2018). Cells were transduced at a multiplicity of infection (MOI) of 0.31 using 10 µg/ml polybrene at 700x coverage/sgRNA. Screen2: validation screen: Cells were transduced at an MOI of 0.1 using 10 µg/ml polybrene with a 500x coverage/sgRNA.
