Extreme, permanently anoxic environments are critical frontiers for discovering novel microbial life. The stratified sediments of Etoliko Lagoon, Greece, represent a largely unexplored ecosystem that has been previously suggested to be a hotspot for "microbial dark matter" (MDM). This study provides the first comprehensive, multi-domain characterization of the bacterial, archaeal, and eukaryotic communities within these sediments, employing a dual short-read (Illumina) and long-read (Nanopore) sequencing approach coupled with phylogenetic analysis.
