Expression and clinical significance of CXCL8 in cervical cancer

1.Bioinformatics analysis based on online databasesBy integrating the data resources of the Cancer Genome Atlas (TCGA) and GTEx databases, we used the biomedical data analysis tool SangerBox to explore in depth the differential expression of the CXCL8 gene in a variety of cancerous tissues and normal tissues. Further, with the help of the GEPIA online database, we analyzed the expression differences of the CXCL8 gene in cervical cancer and its paracancerous tissues in detail. In order to comprehensively assess the impact of CXCL8 gene expression on cancer prognosis, we performed an in-depth analysis of the relationship between the CXCL8 gene and pan-cancer prognosis using overall survival (OS) as an assessment index. Specifically, in cervical cancer (CESC) patients, we further explored the association between CXCL8 gene expression and prognosis. When determining the cutoff value, we strictly based it on the principle of sample size proportion to ensure the rationality and statistical validity of grouping, i.e., the minimum grouping sample size proportion should not be lower than 25%, and the maximum should not be more than 75%. Based on this scientific grouping, we divided the patients into two groups of high and low expression and utilized the survival package and survfit function of R software to conduct a detailed comparative analysis of the prognosis of the two groups.2.Clinical validationA total of 94 patients with cervical squamous cell carcinoma admitted to the First Affiliated Hospital of Xinjiang Medical University between January 2017 and December 2021 were included in this study, all of whom had complete clinical data and pathological tissue samples. The pathological tissues of the above patients were routinely prepared into immunological sections for immunohistochemistry experiments to analyze the expression of CXCL8 and its clinical significance in this population. Statistical analysis was performed using SPSS software. Measurement data were statistically described by mean±standard deviation, count data were statistically described by relative composition ratio/rate, rank data were statistically analyzed using the rank-sum test, and the relationship between the protein expression of CXCL8 and the clinical data of patients with cervical cancer was analyzed using the chi-square or Fisher's exact test. p<0.05 was taken to mean that the difference was statistically significant.
