Vertebrates eDNA samples of the Colombian Amazon and Orinoco basins

Enviromental DNA samples were collected in 29 locations of 4 colombian zones (Northeastern orinoco, southwestern orinoco, northwestern amazonas and southern amazonas) using NatureMetrics eDNA collection kits. After DNA extraction and purification, the hypervariable region of the mitochondrial 12S ribosomal RNA encoding gene was amplified to target fish as part of the ​eDNA survey - Vertebrates​ pipeline. Twelve PCRs replicates were made for each of the 29 filters. The purified index PCRs were pooled into a final library with equal concentrations and then was sequenced using an Illumina MiSeq V3 kit at 12 pM with a 10% PhiX spike.
