Transcriptome and mRNA stability analysis of neuronal differentiation

RNA-seq and mRNA stability assay were used to probe changes occuring neuronal differentiation and maturation of SH-SY5Y cells. We analyzed differentially expressed genes after neuronal maturation via RNA-seq. We also analyzed mRNA stability by utilizing Actinomycin D mediated transcription blockage followed by RNA-seq. Libraries were PolyA enriched for mRNA and prepared using NEBNext Ultra II Directional RNA Library Prep Kit for Illumina.
