4-octyl itaconate regulates immune response of brain immune cells following traumatic brain injury. Overall design: 11-12 weeks old male mice were subjected to sham (n=8) or controlled cortical impact injury (n=20). At 1 hour later, CCI mice were randomly administrated with vehicle (40% cyclodextrin in PBS) (n=10) or 4-octyl itaconate (4-OI, 10 mg/kg) (n=10) via intraperitoneally injection. Mice were sacrificed and perfused 12 hours after CCI and ipsilateral brain hemispheres were collected. CD11b positive cells were isolated through CD11b+-magnet beads. The isolated CD11b positive cells from Sham, TBI+Vehicle, and TBI+4-OI groups were randomly divided into two samples, respectively, for single-cell RNA sequencing analysis.
